چکیده

We have evaluated and modified a current method for estimating 8-aminoIevulinic acid dehydratase (5-aminolaevulinate hydro-lyase, EC 4.2.1.24) activity in blood, so as to adequately control the pH of the enzyme-substrate solution at the optimum throughout the incubation period. This has improved the sensitivity and reliability of the assay. Also, N-ethylmaleimide has been substituted for a potentially hazardous mercury salt, used to remove sulfhydryl groups before color development with a modified Ehrlich’s reagent.

Lead toxicity in the pregnant rat. II. Effects of low-level lead on delta-aminolevulinic acid dehydratase activity in maternal and fetal blood or tissue.

Lead was administered to pregnant rats in drinking water (5 ppm) from day 1 to day 18 or 21 of gestation, at which time the dams and fetuses were sacrificed. The red blood cell count and hemoglobin concentration were significantly lower in the lead-treated fetuses than in untreated controls on day 21 of gestation, but not in the lead-treated dams. The delta-aminolevulinic acid dehydratase (ALAD...

Biological Monitoring Among Workers Exposed to Inorganic Lead and Its Compounds

Material and Methods: We performed cross-sectional study using 60 workers occupationally exposed to lead compared with 60 controls. All examinees were assessed by Questionnaire, and laboratory testing concerning blood lead level (BLL), activity of delta-aminolevulinic acid dehydratase (ALAD) in blood, concentration of delta-aminolevulinic acid (ALA) and coproporphyrin in urine, reticulocytes an...

Induction of delta-aminolevulinic acid dehydratase in mouse Friend virus-transformed erythroleukemia cells during erythroid differentiation.

The activity of delta-aminolevulinic acid (ALA) dehydratase, an enzyme involved in heme biosynthesis, has been shown to increase in Friend virus-transformed murine erythroleukemia (MEL) cells during erythroid differentiation. In this study, the nature of the increase in ALA dehydratase activity in MEL cells was examined using a monospecific antibody directed to the enzyme. A sevenfold increase ...

Delta-aminolevulinic acid dehydratase activity in the general population of southern Minas Gerais, Brazil.

Blood samples were collected from 113 subjects (56 males and 57 females) living in the district of Alfenas, in southern Minas Gerais state, Brazil, to establish reference values for delta-aminolevulinic acid dehydratase activity (ALA-D, EC 4.2.1.24). The state of health of the population was confirmed by hematological and biochemical parameters analyzed in blood and urine samples. ALA-D determi...

Erythrocyte uroporphyrinogen I synthase activity in diagnosis of acute intermittent porphyria.

Measurement of the activity of uroporphyrinogen I synthase provides an excellent laboratory aid in the diagnosis of acute intermittent porphyria, particularly in those patients who are asymptomatic or in whom the disease is not biochemically manifested by porphyrin precursor excretion. We describe here a simplified fluorometric method for measuring the activity of this enzyme in whole blood. Th...
